I have been working some on a load for my bisley in 44.I have a 305wfngc loaded over 21.5gr of H110 in new starline brass with cci 350 primers.I have another load which has a 320wfngc over 20.5gr of H110 in new starline brass with cci 350 primers.Out of my 5 1/2 bl I am guessing 1250-1300fps for the 305 and 1100-1150fps for the 320.So that being said 1.)which should offer the most penetration? 2.)And if shot from a 6 1/2"bl and a 7 1/2"bl would there be any significant penetration differences?

For all practical purposes...none.......take the load that shoots the best and be happy!

If the nose profiles of the two bullets are similar, then I would als say that there is no practical difference.

Sound like you are getting good advice. as Dan has stated, I would go with the one that has the better groups.

Hmmmmm.....set up a 25 yard 3inch paper target.
Pull out both boxes of said loads.
For a pistol rest, stack some sandbags with a piece of leather on top.*
Let your Shooting Buddy who didn't buy(Friday Night at the Bar) a round of Suds, but was more than happy to down the ones everyone else was willing to pay for, take the first shots!
After a dozen rounds and pistol holds together, give the tightwad a break, continue the session!
The one's that don't turn your thumb and index web into a duck fin, producing a decent group wins!

WolfTrap has a great idea -- except stretch the range out to 50 yards! That'll teach him not to buy any rounds!
