Looks can be deceiving but doesn't the case appear a bit larger than a 45?

Rod, too.

I did some calculations based on some rough measurements I took of that picture, and assuming that case takes a large pistol/rifle primer, then that cartridge has a rim diameter of about .566". The closest rim diameter in a handgun cartridge to that is the 500 S&W at .560". But, I could be wrong.

Could it be that .480 folks have been begging for all these years?

Well, a 480/475 has a rim diameter of about .540, so that's my other guess assuming my measurements were slightly off. Not to mention a 480 is a more reasonable guess for a Ruger.

Most of the rumors are that is a 5 shot 480. I hope it is. An expert with "inside" information posted on another forum that it is a 5 shot 454 Casull.

I could be interested in a .480 Ruger.

Okay, so the rumors and the reverse engineered photo give us an idea of the possible calibers, but what kind of SA is it? BH, SBH, Bisley, Bisley Hunter? Is there any possibility it could be a 50 JRH? How about a 50 Ruger? ????????????????

I don't about you guys, but I want some pieces of brass with the Question mark headstamp...it would drive the range officers crazy at indoor ranges who want to "see my ammo"

Any combo mentioned is worth spending a little OT money on. Can't wait to see what it is.
